- Abstract
- Oxidative signaling has important physiological roles in the central nervous system. There is much evidence pointing out the participation of oxidative stress in many pathologic processes such as neuroinflammation, neurodegeneration and cortical brain injury. In astrocytes, oxidative stress is involved in astrogliosis, yielding reactive astrocytes that play roles in glial scar formation, secrete inflammatory cytokines and large amounts of reactive oxygen/nitrogen species. Oxidative stress was also found to influence astrocyte mediated glutamate homeostasis. Specific isoforms of aquaporins were found to play roles in oxidant signaling, but their expression patterns and role in astrocyte remains elusive. We therefore sought to study the role of aquaporin mediated oxidant signaling and stress in astrocytes. We will first assess astrocyte’s expression of specific AQP isoforms suspected to function as peroxiporins, which can transport hydrogen peroxide across membranes. Then, we will measure transport of hydrogen peroxide across membranes of astrocytes. Results could give insight on aquaporin’s roles in oxidative stress and lead to studies aiming at potential disease treatment through intervention on aquaporin’s peroxiporin function.
