Abstract
Nicotiana tabacum commonly known as tobacco serves as a model for studying glandular trichomes development. Trichomes are specialized hair-like structures originating from the epidermis. Tobacco possesses three types of trichomes: non-glandular trichomes, short glandular trichomes, and long glandular ones. The primary role of trichomes is to protect the plant from biotic and abiotic stresses. Additionally, glandular trichomes are of interest due to the compounds they produce, which may be valuable to the pharmaceutical and biotechnology sectors. While the development of non-glandular trichomes is well understood, particularly in Arabidopsis thaliana, the development of glandular trichomes remains less explored. In N. tabacum, the Woolly gene seems to be important for the transcriptional control of glandular trichome development. To gain a deeper understanding of its role, a previous DAP-seq analysis revealed interactions between the NtWoolly transcription factor (TF) and various genes, including the promoters of NtRAX2, NtWoolly, NtZFP8, NtTTG1 and NtGL2. However, DAP-seq results alone are insufficient to confirm these interactions: additional methods are required to validate these findings. As part of this master's thesis, Electrophoretic Mobility Shift Assays (EMSA) and Dual Luciferase Assays were carried out to validate the DAP-seq results. EMSA experiments indicated that Woolly TF binds to its own promoter, the promoter of NtZFP8, and possibly to the promoters of NtTTG1 and NtRAX2. Dual Luciferase Assay experiments demonstrated that Woolly TF acts as a repressor of the NtZFP8 promoter activity and does not significantly impact the activities of other promoters. However, these results provide partial confirmation of the interactions between Woolly TF and the promoters of NtRAX2, NtWoolly, NtZFP8, NtTTG1 and NtGL2. Further experiments are needed to validate these interactions.
